DNA metabarcoding of saproxylic beetles - Streamlining species identification for large-scale forest biomonitoring

Résumé

The PASSIFOR project, presented here, is a 2-year project started in 2013 aiming at developing a DNA metabarcoding approach to enhance and facilitate the use of saproxylic beetles as forest indicators in ecological and conservation biology studies. The objectives of the PASSIFOR project can be divided in two main phases. In Phase 1, we assembled a reference library of 442 DNA barcodes representing 334 species of saproxylic beetles within 32 different families. The PASSIFOR library includes several beetle families so far absent from BOLD or GenBank, and it is expected to stand in the future as a an important reference for DNAbased identification of European beetles, along with complementary iBOL initiatives in other European countries. In Phase 2, we developed and tested DNA metabarcoding protocols to identify species of saproxylic beetles from bulk samples collected in interception traps. The first results of the second phase of the project correlate molecular and morphological identifications of beetles from bulk samples or alcoholic storage fluid only.
